Wissenschaftler aus der Universität Granada, Medical Research Council des Vereinigten Königreichs und Universität Cambridge Sie haben die Tür für die Entwicklung neuer Medikamente gegen neurodegenerative Erkrankungen wie Alzheimer und Parkinson geöffnet.

 

Seine Arbeit, die in der letzten Ausgabe der Zeitschrift Nature veröffentlicht wurde, hat neue Mechanismen entdeckt, die die Erkennung von Polyubikkinketten regeln, ein Protein, das für grundlegende Zellprozesse wie den Abbau von unbrauchbaren Proteinen (Protolyse), Anti-Antikörper-Erkennung, DNA-Transkription und -Reparatur und Zelltod verantwortlich ist.

 

Die Ketten des Ubiquitinproteins wirken in vielen Zellprozessen als Mediatoren, unterstützen den Transport und fördern die Begegnung von Proteinen mit anderen innerhalb der Zelle. Polyubiquitin markiert als "Phoo" Signalisierer jene Proteine, die innerhalb der Zelle nicht mehr nützlich sind und die zerstört werden müssen.

 

Wenn Ubiquitin an das betreffende Protein bindet, identifiziert das Proteosom, das für den Abbau verantwortlich ist, dieses Protein als "abnehmbar" und beginnt eine Kette von Reaktionen, die mit dem vollständigen Abbau des Proteins enden. Die Studie in Nature veröffentlicht zeigt, dass die Identifizierung der Polyubiktinketten, um die Zellfunktion zu beginnen Teil der Auswahl der richtigen Struktur der Kette auf molekularer Ebene.

 

 

Instabile Proteine

 

Die Funktionsstörungen des Polyubikitinkettenregulationssystems sind mit neurodegenerativen Erkrankungen (Alzheimer und Parkinson), dem Angelman-Syndrom oder dem von Hippel-Lindau-Syndrom verbunden. Diese Studie öffnet die Türen für ein besseres Verständnis der Regulierung der Zellfunktionen und Reaktionsmechanismen innerhalb der Zellen gegen das Vorhandensein instabiler Proteine (die zu einer Ansammlung von Faserkörpern in Erkrankungen wie Alzheimer und Parkinson führen können), fremden (viralen) Substanzen und Genomschäden (DNA-Reparatur).

 

Durch die Anwendung von Monomolekular-Flueszenztechniken (eine hochempfindliche Technik, bei der Proteinmoleküle einzeln analysiert werden) weist die Studie auf eine dynamische Vielfalt von Strukturen in den Diubikitinketten (komponiert aus zwei Einheiten des Regulierungsproteins) hin, im Gegensatz zu den statischen Formen, die bisher in den Proteinstrukturen-Repositories festgestellt wurden.

 

Wie die Autoren des Artikels betonen, kann das Verständnis dafür, wie die konforme Auswahl einen ersten Schritt darstellt, der bisher noch nie in der Funktion von Polyubikitis-Ketten nachgewiesen wurde, die Entwicklung neuer Therapien ermöglichen, die auf der molekularen Anerkennung dieser Pathologien basieren.
