Northwestern Medicine In den Vereinigten Staaten, in denen eine biologisch abbaubare Nanopartikel als Transportfahrzeug eines Antigens, das das Immunsystem täuschen, damit es seinen Myelin-Angriff stoppen und ein Modell der rezidivierenden Multiplen Sklerose (MS) beenden kann.
Diese Nanopartikel, die leicht in einem Labor hergestellt und für die Herstellung standardisiert werden können, bestehen aus einem Polymer namens Poly (lactid-co-glykolid) (PLG), bestehend aus einem Milchsäure und Glykolsäure, beide Metaboliten.
In der Multiple Sklerose (EM) von Verletzungen demyelinisierende Stoffe, Neurodegenerative und Chroniken ZentralnervensystemDas Immunsystem greift die Myelinmembran an, die die Nervenzellen im Gehirn, im Rückenmark und im Sehnerv schützt. Sobald diese Isolierung zerstört ist, können die elektrischen Signale nicht effektiv durchgeführt werden, was zu mehr oder weniger Taubheitsgefühl, Lähmung oder Blindheit führt.
Als diese Nanopartikel an Myelin-Antigene gebunden und in die Mäuse injiziert wurden, wurde das Immunsystem wiederhergestellt, ohne dass Myelin als fremder Invasor erkannt wurde und seinen Angriff gestoppt wurde. Die Verwendung dieser Nanopartikel unterdrückt nicht das gesamte Immunsystem wie die heutigen Therapien.

Das Team fügte die Myelin-Nanopartikel hinzu, die intravenös in die Mäuse injiziert wurden. Die Partikel kamen in die Milz, die Blut filtert und hilft dem Körper, das Altern und den Tod der Blutzellen zu beseitigen. Dort wurden die Partikel von Makrophagen verschlungen, eine Art Immunzelle, die dann die Antigene auf ihrer Zelloberfläche zeigt. Das Immunsystem sah Nanopartikel als normale Blutzellen, die starben und sich nicht sorgten, was die Immuntoleranz des Antigens verursachte, indem es direkt die Aktivität der gegen Myelin empfindlichen T-Zellen hemmte und die Anzahl der T-Zellen erhöhte, die Autoimmunreaktion beruhigte.
Zukunft
Diese neue Nanopartikel kann auch auf eine Vielzahl von Autoimmunerkrankungen angewendet werden, indem man einfach das gelieferte Antigen, einschließlich Typ-1-Diabetes, Lebensmittelallergien und Atemwegsallergien wie Asthma ändert.
Die Entwicklung dieses Nanopartikels hat es getan Lonnie Shea, Professor für chemische und biologische Technik an der Fakultät für angewandte Ingenieurwissenschaften Northwestern University in Evanston (Illinois, USA). Die Herstellung erfolgt aus einem bereits von der Amerikanische Arzneimittelagentur (FDA).
Das Ergebnis dieser Studie an Mäusen ist sehr positiv, da es gezeigt hat, dass Nanopartikel ebenso wirksam sein können wie weiße Blutzellen als Lieferwagen des Arzneimittels.
